Identification of a Clinically Efficacious CAR T Cell Subset in Diffuse Large B Cell Lymphoma by Dynamic Multidimensional Single-Cell Profiling

Using infusion products of individuals with large B cell lymphoma, the authors integrated functional profiling using timelapse imaging microscopy in nanowell grids with subcellular profiling and single-cell RNA sequencing to identify a signature of multifunctional CD8+ T cells.

γδ T Cell Profiling in a Cohort of Preterm Infants Reveals Elevated Frequencies of CD83+ γδ T Cells in Sepsis

γδ T cells were investigated in a longitudinal cohort of preterm neonates using next-generation sequencing, flow cytometry, and functional assays. During the first year of life, the Vγ9Vδ2 T cell subset showed dynamic phenotypic changes and elevated levels of fetal-derived Vγ9Vδ2 T cells in infants with sepsis.

Molecular Subgroups of T Cell Acute Lymphoblastic Leukemia in Adults Treated According to Pediatric-Based GMALL Protocols

Researchers presented a homogenously treated cohort of 230 adult T-lineage lymphoblastic leukemia patients characterized on transcriptome, and partly on DNA methylation and gene mutation level in correlation with clinical outcome.

    IL-3-Driven T Cell–Basophil Crosstalk Enhances Antitumor Immunity

Scientists investigated the role of interleukin (IL)-3 in orchestrating anti-tumor immunity through cytotoxic T lymphocyte (CTL) modulation. Intratumoral CTLs underwent a progressive decline in IL-3 production, which was correlated with impaired cytotoxic function.

A Bispecific Antibody Targeting HLA-DQ2.5-Gluten Peptides Potently Blocks Gluten-Specific T Cells Induced by Gluten Ingestion in Patients with Celiac Disease

Investigators assessed the ability of DONQ52 to inhibit coeliac disease patient-derived T cell responses to the most immunogenic gluten peptides encompassing immunodominant T cell epitopes.

Development and Interpretation of a Pathomics-Driven Ensemble Model for Predicting the Response to Immunotherapy in Gastric Cancer

Scientists developed and validated a pathomics-driven ensemble model for predicting the response to immune checkpoint inhibitors in gastric cancer, using H&E-stained whole slide images.

Keratinocytes Present Staphylococcus aureus Enterotoxins and Promote Malignant and Non-Malignant T Cell Proliferation in Cutaneous T Cell Lymphoma

Expression of HLA-DR, high-affinity receptors for Staphylococcal enterotoxins (SE), by keratinocytes correlated with IFN-γ expression in the tumor microenvironment. Importantly, IFN-γ induced HLA-DR, SE-binding, and SE-presentation by keratinocytes to malignant T cells from Sézary syndrome patients.

Bevacizumab Induces Ferroptosis and Enhances CD8+ T Cell Immune Activity in Liver Cancer via Modulating HAT1 and Increasing IL-9

Researchers investigated whether and how bevacizumab regulated ferroptosis and immune activity in liver cancer. They performed RNA-sequencing in bevacizumab-treated human liver cancer cell line HepG2 cells, and found that bevacizumab significantly altered the expression of a number of genes.

Fulminant Hepatitis in a Hepatitis B Surface Antigen-Positive Patient with Adult T-Cell Leukemia-Lymphoma after Mogamulizumab Monotherapy

Scientists reported a case of fulminant hepatitis in a hepatitis B surface antigen-positive patient with aggressive adult T cell leukemia-lymphoma who received monotherapy with an anti-CCR4 monoclonal antibody, mogamulizumab, with decreased hepatitis B virus- DNA levels by entecavir prophylaxis.

Arming Vδ2 T Cells with Chimeric Antigen Receptors to Combat Cancer

The authors present how arming Vδ2-T cells might be the basis for next-generation immunotherapies against solid tumors. They also discuss the clinical potential of Vδ2 CAR-T cells to overcome the current limitations of immunotherapy in solid tumors.
