Epigenetic Therapy Potentiates Transposable Element Transcription to Create Tumor-Enriched Antigens in Glioblastoma Cells

Researchers treated patient-derived primary glioblastoma stem cell lines, an astrocyte cell line, and primary fibroblast cell lines with epigenetic drugs, and identified treatment-induced, transposable element-derived transcripts preferentially expressed in cancer cells.

Novel Arf1 Inhibitors Drive Cancer Stem Cell Aging and Potentiate Anti-Tumor Immunity

Two new potent small-molecule inhibitors of Arf1, identified as DU101 and DU102, for cancer therapy were introduced. Preclinical tumor models demonstrated that these inhibitors triggered a cascade of aging in CSCs and enhanced anti-tumor immunity in mouse cancer and PDX models.

Genetically Engineered CD276-Anchoring Biomimetic Nanovesicles Target Senescent Escaped Tumor Cells to Overcome Chemoresistant and Immunosuppressive Breast Cancer

Investigators developed new approaches for selectively killing senescent-escape CSCs. High CD276 expression in multidrug-resistant breast cancer cells, facilitated immune evasion by low-immunogenic senescent escape CSCs.

The Dual Role of POSTN in Maintaining Glioblastoma Stem Cells and the Immunosuppressive Phenotype of Microglia in Glioblastoma

Scientists investigated the role of POSTN in maintaining glioblastoma stem cells and the immunosuppressive phenotype of microglia.

Identification of SPP1+ Macrophages in Promoting Cancer Stemness via Vitronectin and CCL15 Signals Crosstalk in Liver Cancer

Investigators analyzed single-cell RNA sequencing data from 12 patients with liver cancer and identified a subpopulation of macrophages characterized by elevated expression of SPP1. These SPP1+ macrophages induced upregulation of tumor stemness through a vitronectin-dependent paracrine mechanism.

DRD4 Promotes Chemo-Resistance and Cancer Stem Cell-Like Phenotypes by Mediating the Activation of the Akt/β-Catenin Signaling Axis in Liver Cancer

To investigate the gene and protein expression profiles of DRD4 in liver cancer stem cells (LCSCs) and non-LCSCs, scientists utilized transcriptome sequencing and Western blotting analysis.

FOSL1 Promotes Stem Cell-Like Characteristics and Anoikis Resistance to Facilitate Tumorigenesis and Metastasis in Osteosarcoma by Targeting SOX2

The dynamic expression patterns of Fos‑like antigen 1 (FOSL1) were investigated during the acquisition of cancer stem‑like properties using RNA sequencing, PCR, western blotting, and immunofluorescence.

    Abstract

Acid-Sensing Ion Channel 3 Is a New Potential Therapeutic Target for the Control of Glioblastoma Cancer Stem Cells Growth

Researchers reported that the acid-sensing ion channel 3 isoform, usually absent in the central nervous system and present in the peripheral nervous system, was enriched in human glioblastoma cancer stem cells while poorly expressed in the healthy human brain.

TRIM24/ZFX Affects the Stemness and Resistance to 5-FU of Colorectal Cancer Cells

Scientists explored the effects of tripartite motif containing 24 (TRIM24) and zinc finger protein, X-linked (ZFX) on colorectal cancer cell stemness and 5-FU resistance.

Tools to Study Neural and Glioma Stem Cell Quiescence

The authors discuss the in vitro and in vivo models that have been developed for studying neural stem cell quiescence and how these tools may be used to deepen biological understanding and to develop novel therapies targeting quiescent glioma stem cells.

Understanding the Intersection Between Placental Development and Cancer: Lessons From the Tumor Suppressor BAP1

Investigators explore the multifaceted role of the tumor suppressor BRCA1-associated protein 1 (BAP1) in cancer and placentation. Initially recognized for its tumor-suppressive properties, BAP1 has emerged as a key regulator at the intersection of tumorigenesis and placental development.
