    Targeting
PAK4 to Reprogram the Vascular Microenvironment and Improve CAR-T Immunotherapy for Glioblastoma

PAK4 knockout induced adhesion protein re-expression in endothelial cells, reduced vascular abnormalities, improved T cell infiltration and inhibits glioblastoma growth in mice.

The Vascular Landscape of Human Cancer

Scientists developed a standardized metric- the Endothelial Index- to estimate vascular density in over 10,000 human tumors, corresponding to 31 solid tumor types, from transcriptome data.

The N-Glycome Regulates the Endothelial-to-Hematopoietic Transition

Researchers showed that microRNA-223 (miR-223)–mediated regulation of N-glycan biosynthesis in endothelial cells (ECs) regulated endothelial-to-hematopoietic transition. MiR-223 was enriched in hemogenic endothelial cells and in oligopotent nascent hematopoietic stem and progenitor cells.

Cell Indoleamine 2, 3-Dioxygenase 1 Alters Cardiac Function Following Myocardial Infarction through Kynurenine

Cardiac repair was analyzed in mice harboring specific endothelial or smooth muscle cells or cardiomyocyte or myeloid cell deficiency of indoleamine 2, 3-dioxygenase 1 and challenged with acute myocardial infarction.

Alleviates Hypoxia-Induced Blood-Brain Barrier Injury via Regulation of CLDN5 (Claudin 5)

The authors report that in brain microvascular endothelial cells of patients suffering stroke, CLDN5 abnormally aggregated in the cytosol accompanied by autophagy activation. In vivo zebrafish and in vitro cell studies revealed that blood-brain barrier breakdown was partially caused by caveolin 1-mediated redistribution of membranous CLDN5 into the cytosol under hypoxia.

Inhibits VEGF/VEGFR2 Axis of Choroidal Endothelial Cells and M1-Type Macrophages to Protect against Mouse Laser-Induced Choroidal Neovascularization

Investigators HMPL-013 inhibited VEGF/VEGFR2 binding in choroidal endothelial cells and macrophages, as well as macrophage M1 polarization. In vitro, noncontact coculture of human choroidal vascular endothelial cells and macrophages under hypoxia conditions was established.

Eicosapentaenoic Acid Reduces Prostate Tumor Vascularity

Testing autocrine and paracrine effects in an avascular tumor spheroid growth assay, both exogenous monoacylglyceride-conjugated form of eicosapentaenoic acid and endogenous ω3 reduced VEGF secretion and in vitro endothelial cell tube formation and blocked tumor spheroid growth, suggesting that ω3 molecules could directly hinder prostate cancer cell growth.

Generation of Immune Cell Containing Adipose Organoids for In Vitro
    Analysis of Immune Metabolism

Scientists created the first 3D adipose tissue organoid model which could contain and maintain resident immune cell populations of the stromal vascular fraction and proved to be effective in studying adipose tissue biology in a convenient manner.

Fibroin Vascular Graft: A Promising Tissue-Engineered Scaffold Material for Abdominal Venous System Replacement

The authors focused on the development of new tissue-engineered vascular graft using a silk-based scaffold material for abdominal venous system replacement. A rat vein, the inferior vena cava, was replaced by a silk fibroin tissue-engineered vascular graft.

Mediated Signaling within Arteries Promotes Vascular Smooth Muscle Cell Recruitment

Investigators showed that autocrine cxcl12b/cxcr4 activity led to increased production of the vascular smooth muscle cells chemoattractant ligand pdgfb by endothelial cells in vitro and increased expression of pdgfb by arteries of zebrafish and mice in vivo.
